Xilio Therapeutics is a clinical-stage biotechnology company with a vision to transform the lives of patients with cancer by harnessing the power of highly potent, tumor-selective immuno-oncology (I-O) therapies that deliver deep and durable clinical responses.
Current I-O, therapies have curative potential for patients with cancer; however, their potential is significantly curtailed by systemic toxicity that results from activity of the therapeutic molecule outside the tumor microenvironment (TME).
Our team of experienced scientists and researchers are using our proprietary geographically precise solutions (GPS) platform to rapidly engineer novel molecules, including cytokines and other biologics, that are designed to optimize their therapeutic index. These molecules are engineered to localize activity within the TME without systemic effect, resulting in the potential to achieve enhanced anti-tumor activity.
We are advancing several GPS designed agents through preclinical and clinical development, including XTX101, a tumor-selective anti-CTLA-4 monoclonal antibody, XTX202, a tumor-selective IL-2 as well as XTX301, and a tumor-selective IL-12.

Xilio was founded in 2016 under the name Akrevia Therapeutics. In 2020, the company rebranded to Xilio Therapeutics to reflect the company’s evolution to a development stage company.
